研究の背景と目的 Background and Purpose of Study

細胞は,様々なストレス環境に適応するためにストレス応答システムを持っています。その一つである小胞体ストレス (ERストレス) は,各種ストレスによって構造異常のタンパク質が蓄積した時に誘導され,ERストレス応答 (unfolded protein response, UPR)と呼ばれる細胞の生存のための反応がおこります。このシステムは,タンパク質の品質管理に重要で,異常タンパク質の分解にはプロテアソームやオートファジーが関与します。一方,ストレスが許容範囲を超えると,細胞死が誘導されます。遺伝子組換えやバイオインフォティクスの手法を用いて,ERストレス,ヒートストレス,メカニカルストレス等による細胞応答のメカニズムを解明する研究を行っています。


In order to adapt stressful conditions, cells have developed cellular stress response systems. One of these is endoplasmic reticulum (ER) stress which is defined as accumulation of unfolded proteins. ER stress induces a coordinated cytoprotective program called unfolded protein response (UPR), and is vital to the protein quality control and the proteasome and autophagy play a key role in it. If the stress is beyond capacity of the adaptive machinery, cells undergo cell death. We are currently studying the molecular mechanisms underlying cellular responses to ER stress, heat stress, mechanical stress, etc. using recombinant genetic engineering and bioinformatics technologies.

基本的には,医療応用を目指して研究を行っています.新たな発見を行うためには,様々なアイデアを持った研究者との交流が非常に重要であると考えています.現在,薬学,医学,農学,理学,工学,歯学のバックグラウンドを有する研究者と組織横断的な研究を展開しています.

研究内容

超音波による細胞機能制御に関する研究

超音波は20 kHzを超える周波数の粗密波で,超音波探査や洗浄等,我々の身の回りに幅広く利用されています.また,超音波は医療分野において,診断に必須の技術であるのみならず,癌温熱療法,結石破壊,白内障の手術,血栓溶解,骨折治療等,治療にも利用されています. 低出力パルス超音波 (LIPUS) によるメカニカルストレスが骨折治療に有用なことは良く知られていますが,その分子メカニズムは依然不明な点が多いです.我々は, LIPUSをマウス前骨芽細胞様細胞MC3T3-E1に照射し,マイクロアレイを用いてLIPUSに応答する遺伝子群と遺伝子ネットワークを明らかにしました (1).これは単一の細胞から得られた成績であり,生体におけるLIPUSの効果をより正確に理解するためには,複数の細胞からなる組織やモデル動物で評価することも重要です.魚類のウロコには,ほ乳類の骨と同様に骨芽細胞と破骨細胞が存在し,骨の機能を評価する有用なモデルであることが知られています.LIPUSをウロコモデルに作用させた時,骨芽細胞が活性化し,破骨細胞にアポトーシスを誘導するという成績が得られています (2, 3).現在,細胞,組織(ウロコモデルを含む)と動物モデルを用いて,LIPUSによる細胞機能制御に関する研究を行っています.

ストレス応答の分子メカニズムに関する研究

生体が熱に曝された時(ヒートストレス),細胞の中ではheat shock response (HSR) が起こり,直ちに転写因子heat shock transcription factor 1 (HSF1) が活性化され,heat shock proteins (HSPs) が発現します.HSF1とHSP関連タンパク質の細胞機能における役割やそれらとがんとの関連性に注目しています (4-7). 地球環境中には非常に沢山の化学物質が存在し,生物に様々な影響をおよぼしています.最近,フッ化ナトリウムが細胞にERストレスを伴う細胞死を誘導すること (8) や魚類の骨関連の細胞に影響を与えてCa2+濃度を上昇させることを明らかにしました (9).
