研究の背景と目的 Background and Purpose of Study

今までに,外側手綱核(Lateral habenula: LHb)は,幼少期にストレス感受性が高く(生後10-20日),この時期にストレスを受けたマウスでは,成長後,LHbにおいてParvalbumin(PV)陽性神経細胞数が少なく,ストレス刺激後に活動した神経細胞数が多く(PV陽性神経細胞とは異なる細胞),不安・うつ様行動を呈することを明らかにしました.LHbは経験に依存して成熟し,その成熟障害が不安・うつ様行動発症に関わることが示唆されます.経験に依存して変化するLHbの神経回路を検討し,不安・うつの新規メカニズムを解明します.
We have shown that the lateral habenula (LHb) is highly stress-sensitive during early-life in mice (postnatal day 10-20 ), and that mice exposed to stress during this period have fewer Parvalbumin (PV)-positive neurons in the LHb and abnormally high stress reactivity (different from PV-positive neurons) and exhibit anxious and depressive-like behaviors. The results suggest that experience in early life are essential for the maturation of LHb that underlie emotion. We will examine the neural circuitry of experience-dependent changes in LHb and elucidate a novel mechanism of anxiety and depression.  

本研究の領域横断性

マウスの脳の染色(免疫染色,in-situ HCR),行動実験(行動解析スクリプト),脳波の解析スクリプトの作成で共同研究可能です. 特にマウスの脳の染色では,神経細胞活動性マーカーを詳細に解析することで,新規脳部位を発見したという実績があります(Kanemoto et al., 2020)

研究内容

今までに以下のことを明らかにしています.
  • 情動を司る外側手綱核という脳部位の神経回路が4つの段階を経て、成熟することを明らかにしました。
  • 外側手綱核が未熟な時期(生後10-20日:マウスは生後20日程度で離乳)にストレスを与えたマウスでは、神経回路の成熟の変容と、成長後の不安とうつ様行動を呈しました。
  • 外側手綱核が一定の幼少時期(生後10-20日)に回路の変容が起こりやすく、この時期の経験が情動の成熟に重要であることを示しました。この回路変化は、不安とうつが発症する脳内機序であると考えられます。また、幼少の特定の時期の経験が生涯に渡る不安とうつを予防するのに非常に重要であることが示唆されます。本研究は、不安とうつの治療と予防、幼少期子育ての効果の研究基盤となります.
